Research Areas

1) The mechanism of dopamine biosynthesis and catabolism
■ L-DOPA-유도 dopamine 생합성 작용 및 대사과정 작용기전: L-DOPA를 PC12 세포내에 가하면 세포내 TH 활성 및 TH mRNA 함량 증가에 의하여 dopamine 함량이 증가하며, 작용기전인 cyclic AMP-PKA-CREB 신호전달계 및 Ca++-PKC-CaMK II 신호전달계에 대하여 고찰함.
■ Dopamine 생합성 제어제 도출: Scoparone, Seasamin, HK006는 PC12 세포내 dopamine 함량 증가작용을 나타내며, 작용기전 cyclic AMP-PKA-CREB system에 대하여 고찰함.


2) Neurodegenerative diseases and oxidative stress
■ 산화 스트레스(oxidative stress)-유도 신경세포 독성 신호전이: L-DOPA/6-OHDA/metals 등은 PC12 세포에서 산화 스트레스에 의하여 세포사(apoptosis)를 나타내며, 이 과정에서 cyclic AMP 함량이 증가함. 산화스트레스에 의한 세포사(survival/death) 신호전이에 대하여 고찰함.
■ 신경퇴행성 질환 동물모델 이용 후보약물 도출 및 작용기전: 뇌신경 세포사(dopamine 신경세포)-유도 동물모델(파킨슨병 모델)을 작성하고 후보약물(RCBH, HK006, scoparone 등)을 도출하고 이의 작용기전을 고찰함.

3) Protective agents for neurodegenerative diseases

■ Neurite outgrowth-유도작용 후보약물의 활성검색 및 작용기전: Neurite outgrowth 유도작용 후보약물의 활성검색 및 작용기전/신호전이 기전을 규명하기 위하여 neurite outgrowth 측정, cyclic AMP 함량, ERK/PKA/PKC/CaMK II/MAPK 활성을 측정함.
■ 신경전달물질 유리(dopamine release) 유도작용 제어제: Scoparone은 PC12 세포모델에서 dopamine 유리작용을 나타내며, 이의 작용기전은 cyclic AMP-Ca++ influx-PKA-CaMK II- p-synapsin 활성이 관여하고 있음.
